Jacob and Matt Vinchest they are brothers with hard fate. When they were young, their mother Mary died a violent and unexplainable death, which in turn led their father, John Winchest, to teach them skills which they might need in the search for whatever it was that took her life.

Subsequently, he raised them to be soldiers. He taught them about the paranormal evil that lives in the dark corners and he taught them how to kill it.

But Jacob didn’t want such lifestyle. He left home to begin "normal" life.

Years later, while attending Stanford University, Jacob is forced back into the paranormal world by his older brother, who has come to tell him that their father has mysteriously disappeared.

Jacob later returns home, and discovers his girlfriend Jessica pinned to the ceiling. He is then forced to watch as she is killed in the same supernatural manner as his mother. Jessica's death prompts him to return to the life of hunting with Matt.

The brothers set out on a journey, fighting evil along the way, to find their recently missing father who, when they finally meet up with, reveals he knows what killed their mother, a demon, and have found a way to track and kill it.

Along the way, Jacob and Matt save innocent people, fight creatures and ghosts, and collect clues to their father's whereabouts. They eventually find their father, who reveals that the creature that killed Jacob and Matt 's mother years earlier is Yellow-eyes (Azazel) and the only thing that can kill him is a legendary gun created by Samuel Colt.

  

Demon order to force them to hand over the Colt, but they refused.

However, Azazel soon reveals himself to be in possession of John, and attacks them. As the Azazel tortures Matt, though, John is able to temporarily gain control, and begs Jacob to use the Colt to kill Azazel. Not able to bring himself to kill his father, Sam instead shoots him in the leg, and Azazel flees. On the way to the hospital, the three then crashed into a truck driver.

In the aftermath of the car crash, the surviving Winchesters go to the hospital, where one is stalked by a Reaper while another must consider sacrificing himself to save the others.
